Background:The purpose of the study was to compare the effects of celecoxib and non-steroidal anti-inflammatory drugs (NSAIDs) on the levels of prostaglandin-L-arginine and prostacyclin in patients with osteoarthritis (OA).

Methods:A randomized, open-label crossover study was performed with a double-blinded, randomized, parallel design.

Results:There were no significant differences between the two groups regarding the levels of prostaglandin-L-arginine and prostacyclin in the patients with OA.

Conclusions:The NSAID use was more beneficial for the patients with OA than the other NSAIDs.

Acyclovir (10 mg) and Celebrex (100 mg) In the two-week study, all the patients with OA experienced significantly lower levels of prostaglandin-L-arginine and prostacyclin compared with patients with other types of OA. NSAIDs have been reported to significantly improve pain, inflammation, and stiffness in OA patients, although the effect of NSAIDs is not well-established in clinical practice.Acyclovir and Celebrex In the two-week study, all the patients with OA experienced significantly lower levels of prostaglandin-L-arginine and prostacyclin compared with patients with other types of OA.

Conclusion:In conclusion, the use of non-steroidal anti-inflammatory drugs (NSAIDs) was associated with significantly lower levels of prostaglandin-L-arginine and prostacyclin in patients with OA.

This study was funded by the National Health and Medical Research Council (NHMRC) of the Republic of South Africa, under the Clinical Research Programme Grant No. 6058/B/100/01/09/ and Clinical Research Programme Grant No. 6058/B/100/05/05.

Acute ischemic heart disease (AHD) is a major cause of morbidity and mortality in the worldwide population. It is a major risk factor for cardiovascular diseases, such as coronary artery disease (CAD), hypertension, and cerebrovascular accidents (CVA). Although there is no cure for this disease, the combination of symptomatic and asymptomatic reduction in the risk of stroke and transient ischemic attack has shown to be an effective treatment for AHD. However, recent studies have demonstrated that the use of NSAIDs in patients with AHD is associated with an increased risk of cardiovascular disease. Therefore, a large-scale prospective study was conducted to compare the effects of celecoxib and non-steroidal anti-inflammatory drugs (NSAIDs) on the levels of prostaglandin-L-arginine and prostacyclin in patients with OA.

In the study, a total of 973 patients with OA were enrolled from July 2009 to May 2012 at the University of Pretoria and the South African National Health and Medical Research Council (NHMRC) Research Centre. The patients were randomly allocated to receive either of the following groups: Celecoxib, Celebrex, or the non-selective NSAIDs: Placebo, 200 mg daily (NSAID group), and 200 mg daily (non-selective NSAID group). Patients with an AHD or who were hospitalized within 6 months before the study were excluded. The study was conducted in accordance with the Declaration of Helsinki and Good Clinical Practice guidelines.

Among the 973 patients with OA, the mean age was 37.5 ± 7.6 years. The average time to the onset of the pain in patients with OA was 52.8 ± 9.8 min (range: 21 to 81 min). Celecoxib was administered as a single daily dose and 200 mg as a single daily dose. The mean maximum dose of NSAIDs was 200 mg daily. The mean maximum plasma concentrations of NSAIDs were significantly higher in patients with OA than those with other types of OA (p<0.05). There were no statistically significant differences in the levels of prostaglandin-L-arginine and prostacyclin between the two groups.
